Yes they can be refilled. The biggest problem you might run into is getting the new filler, spackling paste or color putty, to stick. I recommend using a finish nail to dig out some of the filler then refill. This is especially true for color putties used on stained wood.
Yes. Most nail fillers will even re-assure of this on the back label as well. Same goes for caulk
