30 year old home..new trim. Does it have to be primed? I have many partial cans of Semi-gloss off white paint. Will painting it with 2 coats be as good as priming and painting?

Simply? No. A finish coat as a primer coat is not going to give you your best result. Prime one coat and apply two finish coats for the professional result.
